This might be an anti virus software (like MS defender) blocking the downloaded RE from being accessed. That's because new REs are quite often misidentified as malware or suspicious (due to not being whitelisted). Check if there is something in the logs/quarantine of the AV. It's also a good idea to put the RE/VST folders into a "do not scan" exception list to speed up Reason launch.

It's never a good idea to use directory white lists with anti virus software, try disabling while working from known good sources and reenabling once complete.

Having whitelist / do not scan locations will just make the directory a malware target!

Sure in theory, but 1st thing: If you use the Brain 1.0 AV you're pretty safe from malware in the first place:
So things like "not using email on your audio PC", "having an up-to-date locked-down (and probably sandboxed) browser", "having windows show all file extensions", or "running all installer downloads through virustotal.com", you basically don't need any locally installed AV at all. But then having it installed to catch something that might be hiding on a USB drive gives some additional safety net.
Excluding the VST or RE folders is really not much of an issue as malware will first hit in the downloads folder and then in some temp directory and so far I've not heard of any malware that targets vst or RE folders to hide.
Also if the malware is able to install into any of those folders it already has to be running and then it's "game over" anyway as this means your AV was not able to stop it from running in the first place.
