I have a problem in Reason 13.1 on windows with my last song. The song file is 1.2 GB in size, that can't be correct.
In comparison, the previous versions of this song from a month ago are only 120 MB in size.
I've already tried saving the song with "save and optimize", but unfortunately that doesn't help.
I've also checked whether any large samples are embedded, but that doesn't seem to be the case either.
Has anyone had a similar problem and can give me some advice?

Bitley, which optimize in the file menu do you mean, Save and Optimize or the Sample Contain Setting? The "Save and Optimize" doesn't help, the song file is still 1,2 GB big.
The "Song Self-Contain Setting" doesn't help either. It shows that the contained samples are only 4,15 MB big.
Maybe a bug in Reason 13.1?

You may already know this, but if you've been chopping/resizing audio or using comp edit mode, you could still have unused audio accessible in your song file (hidden in resized clips or alternate takes).
Save & Optimize won't delete those for you. You first need to "Bounce Clips to New Recordings" (and Delete Unused Recordings in the comp editor). And then you can Save & Optimize to "defragment" the space you freed up.

I'm having the same issue. Some of my project files are over 1.5 gb and I don't have any self contained samples. I've tried save & optimize a few times and it hasn't helped. I'd love to help diagnose this issue with the devs if possible, it's not sustainable.

The problem is, that I doen‘t have any big audio recordings or audiofiles in the song (see my screenshot above). So this doesn‘t work for me.

I have this issue on Reason 12 even on songs without audio files---the file size often seems to roughly double each time a file saves.

Do your files have audio tracks in the sequencer? To debug it you could create a copy of the file and start to remove elements of the track and then save under a new name to check if it got slimmed down.

I think I found the issue in my project files. I had a combinator patch I made with a few utilities in my Master Section's insert fx that was causing the files to balloon in size. I found this by duplicating my 1.5gb file and deleting tracks one by one, then running save & optimize after each time. None of the tracks in the sequencer made a difference until I deleted the combi on the master and the size reduced from 1.5gb to 51 mb and has stayed around there between subsequent saves and edits. Sounds like a potential bug, but could have also been a third party plugin behaving badly between saves.
